AdEasy™ XL simplifies adenovirus production The gene of interest or expression cassette is first cloned into a shuttle vector (either pShuttle or pShuttle-CMV). The resulting plasmid undergoes a single plasmid transformation into BJ5183-pAD-1. Recombinant adenoviral plasmids are selected on kanamycin and confirmed by restriction digest. The recombinant adenoviral plasmid is digested with Pac I and transfected into AD-293 cells where it replicates and is packaged into virus particles.
Improve Recombinant Adenoviral Production with BJ1583-pAD-1 We compared the capability of BJ5183 and the improved BJ5183-pAD-1 to generate recombinant adenovirus. As shown in the table, generating recombinant adenovirus vector using the original AdEasy system resulted in an 18 to 34% success rate, while the pre-transformed BJ5183-AD-1 resulted in consistently >94% success rate.
