Restriction-Minus Competent Cells for Most Eukaryotic DNA [RUO]

メチル化DNAのクローニング

真核生物のゲノムDNAは高度にメチル化されており、そのメチル化のパターンは組織や発生段階などにより様々です。また、方向性を持たせたcDNAを合成する際、内部の制限酵素サイトがその後のプロセスで切断されないように、メチル化を入れてcDNAを合成することで保護する場合もあります。クローニングされるDNAがE. coli ホストと異なるパターンでメチル化されていると、E. coli ホストの制限システムによって切断され、発現量が著しく偏ることになります。

大腸菌K12株で既知の制限システムをすべて欠失させたこれらのコンピテントセルであれば上記のような切断は受けないので、メチル化されたDNAもクローニング可能です。

BAC クローニング

XL1-Blue MR 細胞は E. coli K12 の既知の制限システムをすべて欠失しています。mcrA、 mcrCB および mrr 突然変異により、シトシンおよび/またはアデニンがメチル化されているDNAの切断が阻止されます。この菌株はまた、F- でありBACクローニングに用いることもできます。
